A RPHPLC(Reverse Phase High Performance Liquid Chromatography) method was found which can determine five βlactam antibiotics under the same conditions simultaneously. The five drugs of cephradine, cephalexin, cefaclor, ampicillin and amoxicillin were determined, using C8 column and HACNaAC buffer (pH value is 3.6)methanol(75∶25) as mobile phase, the UV detector were set at 254 nm. The five drugs can be separated each other in 20min. The detect limits of five drugs were 0.18, 0.21, 0.12, 0.35, 0.25 μg/mL respectively. The proposed method was applied to determine the amount of drugs in human serum and urine and the result was satisfactory. The recovery of five drugs in human serum was all greater than 94.6%, in urine was all greater than 88.3%. It is proved that the method was simple, fast and sensitive.
